Isn't the E.1 one off the first designed to be fighters ?
at least this one is the first with guns that fire through the propellor

The Fokker E.1 was the first aircraft that used an interupter gear to fire through the propeller.  The first purpose designed fighter is probably the Caproni Ca 20.  This nifty one-of-a-kind plane is here in Seattle at the Museum of Flight.
