It depends. The game is loaded from any drive to memory but if there's not enough RAM Windows compensates it by putting some stuff to a cache which usually is in the primary hard disk. Although SSDs are fast, RAM is still faster so if parts of the game have been moved to the cache that will cause hiccups.
